How NMN actually works

NMN sits one enzymatic step from NAD+ — NMN adenylyltransferase converts it directly — which is the basis for the claim that it's the more efficient precursor. The genuine scientific argument is whether it enters cells intact at all: NMN is phosphorylated and therefore not obviously membrane-permeable, and much evidence suggests it's dephosphorylated to nicotinamide riboside outside the cell, absorbed, then re-phosphorylated inside. A dedicated NMN transporter has been proposed but remains debated. If the conversion route is real, NMN and NR are closer to the same product than the marketing suggests.

How to read these tiers

These tiers tell you how much human evidence exists — not how well something works. This is the research space, and most of what’s in here is new rather than disproven. Something sitting at “theoretical” usually means nobody has funded the trial, not that the trial was run and failed.

The trap runs the other way too: something can be clinically validated and still do very little for you specifically. A statistically significant result in a study population is not a promise about your body.

✗ is a safety flag, not a grade. Where you see it, the concern is harm — not a disappointing trial. A compound tested for one purpose and found not to help there can still be worth studying somewhere else, so a negative result never gets rendered as a cross. It sits alongside the tier, because something can be both well-studied and genuinely risky.

Treat it as roughly interchangeable with NR until the transporter question is settled, and buy on price and purity rather than on the step-count argument. Worth knowing that its regulatory status as a supplement in the US has been contested, which affects availability and makes third-party testing more important than usual here. Same caution around active malignancy as NR.
