Nitrosigine
Best-in-class: Nitrosigine
A patented arginine-silicate complex that boosts nitric oxide for pumps, blood flow and even focus — longer-lasting than plain arginine.
How Nitrosigine actually works
Inositol-stabilised arginine silicate. The silicate complex protects arginine from the arginase degradation that limits ordinary arginine, so plasma levels rise higher and stay elevated for hours rather than minutes. Human trials show sustained increases in blood flow, and separately improvements in cognitive measures under fatigue — which fits, since cerebral perfusion responds to the same signal.
Nitrosigine quick facts
| Suggested dose | 1,500 mg pre-workout. |
| How often | Per training session |
| Who it's for | Pumps, blood flow and pre-workout focus. |
✅ Clinically validated
- RCTs show increased blood flow/nitric oxide, better pumps, and some cognitive/focus benefits pre-workout.
- More sustained NO elevation than standard arginine.
📊 Correlative data
- No meaningful observational or traditional-use literature — a synthesised or isolated compound whose entire evidence base is trials and mechanism. Worth stating rather than leaving blank: it means there is no population-level signal either supporting or contradicting what the trials show.
🧪 Theoretical / extrapolated benefits
- Bonded arginine silicate is predicted to raise plasma arginine more durably than arginine alone, and therefore nitric oxide availability — the pitch being sustained rather than acute vasodilation.
- Worth noting that citrulline raises plasma arginine more effectively than oral arginine does, because arginine is heavily metabolised on first pass. The mechanism predicts citrulline should outperform this, and the head-to-head data to settle it does not exist.
These tiers tell you how much human evidence exists — not how well something works. This is the research space, and most of what’s in here is new rather than disproven. Something sitting at “theoretical” usually means nobody has funded the trial, not that the trial was run and failed.
The trap runs the other way too: something can be clinically validated and still do very little for you specifically. A statistically significant result in a study population is not a promise about your body.
- ✅ Clinically validated — human randomised trials or meta-analyses support it. The strongest footing available.
- 📊 Correlative — observational or epidemiological data. Suggestive, and genuinely useful for direction, but it cannot establish cause.
- 🧪 Theoretical / mechanistic — the mechanism is understood and often demonstrated in cells or animals, and the human trial doesn’t exist yet. Unproven is not the same as ineffective. Plenty of what’s standard practice today sat here five years ago.
✗ is a safety flag, not a grade. Where you see it, the concern is harm — not a disappointing trial. A compound tested for one purpose and found not to help there can still be worth studying somewhere else, so a negative result never gets rendered as a cross. It sits alongside the tier, because something can be both well-studied and genuinely risky.
My job is to tell you which one you’re looking at, and let you make the call. Grading something low isn’t me dismissing it — it’s me refusing to oversell it. This is the research space, and being able to reason forward from a mechanism matters as much as waiting for the trial.
A legitimate answer to arginine's bioavailability problem, with actual human pharmacokinetic data behind the specific ingredient rather than the amino acid in general. Effects build over a week or two of daily use rather than being purely acute. More expensive than citrulline for a broadly similar endpoint; the cognitive data is the differentiator if that matters to you.
Nitrosigine — safety & side effects
- Well tolerated; mild GI upset and headache.
- Lowers blood pressure — additive with nitrates and PDE5 inhibitors.
- It is arginine silicate; the silicon content is well below any concern. No significant contraindications in healthy adults.
