How L-Tryptophan actually works

The essential amino acid precursor to serotonin and, downstream, melatonin. The bottleneck is transport rather than supply: tryptophan competes with the other large neutral amino acids for the same blood-brain barrier carrier, and since it is the least abundant of them it usually loses. That is the mechanistic reason a carbohydrate snack raises brain tryptophan — insulin clears the competing amino acids into muscle.

How to read these tiers

These tiers tell you how much human evidence exists — not how well something works. This is the research space, and most of what’s in here is new rather than disproven. Something sitting at “theoretical” usually means nobody has funded the trial, not that the trial was run and failed.

It is a gentler intervention than 5-HTP because the rate-limiting enzyme downstream still regulates conversion, which means less peripheral serotonin and less nausea. The serotonin syndrome interaction with SSRIs, MAOIs and tramadol is real and applies to both.
