How Biotin actually works

Biotin is the covalently-bound cofactor for five carboxylase enzymes — acetyl-CoA carboxylase in fatty acid synthesis, pyruvate carboxylase in gluconeogenesis, propionyl-CoA carboxylase, and the others in amino acid catabolism. It also has a role in keratin production, which is the entire basis of the hair and nail marketing. True dietary deficiency is rare because gut bacteria produce it.

How to read these tiers

These tiers tell you how much human evidence exists — not how well something works. This is the research space, and most of what’s in here is new rather than disproven. Something sitting at “theoretical” usually means nobody has funded the trial, not that the trial was run and failed.

The trap runs the other way too: something can be clinically validated and still do very little for you specifically. A statistically significant result in a study population is not a promise about your body.

The clinically important issue is not the supplement's effect on you but its effect on your labs: high-dose biotin interferes with a wide range of streptavidin-biotin immunoassays, and has caused falsely normal troponin results in real heart attacks and falsely abnormal thyroid panels. Stop it several days before any blood test and tell whoever is ordering it.
