L-5-HTP
Best-in-class: 5-Hydroxytryptophan
Direct serotonin precursor derived from Griffonia seed — one step closer to serotonin than tryptophan.
How L-5-HTP actually works
5-hydroxytryptophan sits one enzymatic step from serotonin, downstream of the rate-limiting tryptophan hydroxylase — which is precisely why it works and why it needs care. Bypassing the regulated step means conversion is no longer controlled, and because aromatic amino acid decarboxylase is present throughout the body, much of the conversion happens peripherally rather than in the brain.
L-5-HTP quick facts
| Suggested dose | 50–100 mg, often at night. Start low. |
| How often | Daily |
| Who it's for | Mood, appetite/craving, and sleep-onset support. |
✅ Clinically validated
- Small RCTs suggest benefit for mood and appetite regulation; evidence is promising but smaller than for pharmaceuticals.
- Studied for reducing sleep latency and appetite/craving.
📊 Correlative data
- Low serotonergic tone is associated with low mood, carbohydrate craving and poor sleep.
🧪 Theoretical / extrapolated benefits
- Proposed to raise central serotonin; the concern is unproven long-term effects and the theoretical need to pair with cofactors.
These tiers tell you how much human evidence exists — not how well something works. This is the research space, and most of what’s in here is new rather than disproven. Something sitting at “theoretical” usually means nobody has funded the trial, not that the trial was run and failed.
The trap runs the other way too: something can be clinically validated and still do very little for you specifically. A statistically significant result in a study population is not a promise about your body.
- ✅ Clinically validated — human randomised trials or meta-analyses support it. The strongest footing available.
- 📊 Correlative — observational or epidemiological data. Suggestive, and genuinely useful for direction, but it cannot establish cause.
- 🧪 Theoretical / mechanistic — the mechanism is understood and often demonstrated in cells or animals, and the human trial doesn’t exist yet. Unproven is not the same as ineffective. Plenty of what’s standard practice today sat here five years ago.
✗ is a safety flag, not a grade. Where you see it, the concern is harm — not a disappointing trial. A compound tested for one purpose and found not to help there can still be worth studying somewhere else, so a negative result never gets rendered as a cross. It sits alongside the tier, because something can be both well-studied and genuinely risky.
My job is to tell you which one you’re looking at, and let you make the call. Grading something low isn’t me dismissing it — it’s me refusing to oversell it. This is the research space, and being able to reason forward from a mechanism matters as much as waiting for the trial.
The interaction is the headline and it is not theoretical: combined with an SSRI, SNRI, MAOI, tramadol or triptan it can cause serotonin syndrome, which is occasionally fatal. Peripheral conversion also causes the nausea people report, and is why research settings pair it with carbidopa. Vitamin B6 is required for the conversion. Effective for some, and it demands respect rather than casual use.
L-5-HTP — safety & side effects
- Nausea is very common and dose-limiting; taking it with food and starting low helps. Vivid dreams and morning grogginess.
- Do not combine with SSRIs, SNRIs, MAOIs, triptans or tramadol without medical supervision. Serotonin syndrome is rare but it is a genuine emergency — agitation, tremor, fever, rigidity — and the risk is additive rather than threshold-based. 5-HTP is the most likely thing in this catalogue to cause it, because it directly supplies the substrate.
- Do not combine with carbidopa outside medical supervision. Long-term safety has not been characterised — the trials run weeks to months, not years. That is a real limit on what anyone can tell you about daily use for a decade.
Not medical advice. If you take prescription medication or have a diagnosed condition, check this against it with a pharmacist or doctor — pharmacists are underused and free.
