The Khavinson literature behind this is decades deep, almost entirely Russian-language, and rarely replicated by any independent group — small single-arm series rather than controlled trials. Absence of replication is not evidence it fails; it is an absence of the evidence that would settle it either way. Dose and course length follow the manufacturer's convention, not a trial. Parathyroid-directed, so calcium handling is the system it sits in — a narrower target than most of the set. Course pattern: roughly 10 days on, then off, once or twice a year. To know whether it did anything, calcium and PTH together, because either alone is uninterpretable. Run it as an experiment you measure, not a protocol you trust.
What Bonothyrk actually is — and why that changes the mechanism
Bonothyrk is a parathyroid-derived peptide complex in capsules, listed as A-21. Of all 13 pages in this cohort it is the one where the standard marketing verb breaks down completely, and the reason is worth more than any hedged paragraph about limited evidence.
'Support' is not a direction, and this gland only has directions. The parathyroids do one job: they hold serum calcium in a narrow band by secreting parathyroid hormone, which pulls calcium from bone, reclaims it in the kidney and activates vitamin D. PTH runs 15-65 pg/mL and calcium 8.6-10.2 mg/dL, and the clinically important states are defined by the COMBINATION. High PTH with high calcium is primary hyperparathyroidism, a condition treated surgically. High PTH with normal or low calcium is usually a normal response to vitamin D deficiency. Low PTH with low calcium is hypoparathyroidism, treated by replacing what is missing. Those are 3 different diseases with 3 different treatments, and 'supporting the parathyroid' points at none of them.
Why that is a mechanism problem and not a wording problem. The class hypothesis is that an organ's own short peptides restore that organ's transcriptional program toward a younger pattern. For most tissues you can at least imagine what 'younger' means. For a gland whose entire function is a set-point controller, 'younger' has to mean tighter regulation around the same set point — which is a testable idea, and a far more specific one than anything on the label, and 0 published experiments have tested it.
What the complex itself commits to, in numbers. A 2-residue peptide has 400 possible identities and a 3-residue one 8,000; the phrase on the label selects none. The 2021 gene-expression review that carries this class's mechanism is indexed by sequence — 98 genes credited to AEDG, 36 to Lys-Glu — and contains 0 parathyroid entries.

Why the Bonothyrk evidence is weak — and what it still showed
Almost every human result in this class comes from one school — Vladimir Khavinson's institute in St Petersburg and the groups around it. That means single-center data, collected by the people who developed the compound, rarely blinded, never pre-registered, and reported across enough endpoints that something was always going to move. Read anything below against that.
Specific to Bonothyrk. The weakness specific to Bonothyrk is conceptual before it is evidential. Parathyroid hormone runs 15-65 pg/mL and calcium 8.6-10.2 mg/dL, and they are interpreted together because the clinically important states are defined by the combination: high PTH with high calcium is primary hyperparathyroidism, a surgical diagnosis; high PTH with normal or low calcium is usually secondary to vitamin D deficiency; low PTH with low calcium is hypoparathyroidism. 'Supporting the parathyroid' does not name a direction, and the directions have opposite treatments. On top of that sits the ordinary gap: 0 published PTH or calcium results for this product, in any species.

Why this particular zero is the most surprising one in the cohort. Several compounds here are aimed at endpoints that are genuinely hard to measure. This one is aimed at 2 numbers that appear on routine panels: PTH at 15-65 pg/mL and calcium at 8.6-10.2 mg/dL. They are cheap, standardized, drawn together as a matter of course, and interpreted as a pair by every clinician who orders them. A single-arm before-and-after in a handful of people would be a weak study and would still be the first data of any kind about this product.
What is cited here and why. The 266-subject elderly series used thymus and pineal preparations by injection over 6-8 years and reported no calcium endpoint, no PTH, no bone density. The 2021 gene-expression review indexes named sequences and has no parathyroid entry. The 2021 independent systematic review pooled 24 randomized trials over 2,245 participants, graded risk of bias moderate to high and certainty of evidence low to very low, and studied cognition. Three real papers, 0 of them about this gland.
What is actually measured, and what is not. Measured: nothing, in 0 published studies. Not measured, and they appear together on a routine panel: PTH at 15-65 pg/mL with calcium at 8.6-10.2 mg/dL, which are interpreted as a pair and define 3 distinct clinical states between them. Also not measured: composition, oral bioavailability, whether anything reaches 4 glands weighing a fraction of a gram each, and the direction the product is even claiming to move.
Not proven is not the same as disproven. Everything above says the evidence is weak. None of it says the compound does nothing. There is no adequately powered trial that ran and came back null, because outside Russia there is essentially no trial at all — this class is unfunded, not failed. A reader who leaves thinking “disproven” has learned something false, and so has one who leaves thinking “proven”.
Bonothyrk pharmacokinetics — how much of it actually gets in
The half-life field reads 'not characterized', which is accurate. A mixture has one clearance curve per component and 0 measurements exist for this preparation in any species.
What can be reasoned, and one genuinely useful comparison. Short peptides reaching plasma meet serum aminopeptidases and clear in minutes. That is worth holding next to the hormone this gland actually makes: intact PTH itself is cleared from the circulation within minutes, which is exactly why intraoperative PTH monitoring works as a real-time surgical test. So a short half-life is not evidence against a peptide doing something here — the native signal in this system is also short-lived. It is evidence that measuring the product in serum would answer nothing, and that the endpoint has to be the regulated variable rather than the regulator.
The oral barrier, unchanged from the rest of the cohort. A swallowed peptide meets gastric acid, pancreatic proteases and then the brush border of the small intestine, where PEPT1 carries di- and tripeptides — 2 and 3 residues — and not complexes; whatever crosses faces hepatic first-pass extraction. 0 oral bioavailability figures have been published for any product in this family, and an injection is 100% bioavailable by definition.
Now do the arithmetic the blank was hiding. Compare the oral products in this class against the injectable ones and the oral form carries roughly 29x more material per day, and on the order of 571x more across a full course. Take an injection as fully bioavailable — 100% by definition, no gut wall, no hepatic first-pass — and the implication is direct: for the oral route to deliver comparable systemic exposure, on the order of 0.2% of what is swallowed would have to arrive in the circulation intact. Whether a peptide mixture can manage that has never been measured — not for this product and not for any product in this family. Stating the bound is honest. Claiming the fraction would not be.
What would have to be true for Bonothyrk to work
The chain. (1) The capsule would have to contain active peptides — 0 published assays. (2) A fraction would have to cross the gut wall intact — the named transporter carries 2- and 3-residue peptides. (3) It would have to reach parathyroid chief cells — never measured; these are 4 glands weighing a fraction of a gram each. (4) It would have to change transcription there — never observed. (5) The regulated variable would have to move in a stated direction — and the direction has never been stated.
The specific test this page insists on. PTH and calcium, drawn together, before and after. Either number alone is close to uninterpretable: a PTH of 70 pg/mL means something completely different with a calcium of 10.4 than with a calcium of 8.8. Anyone running this product who measures only PTH has generated a number that cannot be read, and that is the most common way this gland is misunderstood.
- Prediction 1 — Parathyroid Hormone & Calcium. must be read as a pair — PTH 15-65 pg/mL with calcium 8.6-10.2 mg/dL, before, and annually or after correcting vitamin D. This is the only prediction on the page that is really one test, and it is the whole diagnostic. Either number alone is close to uninterpretable. If a product claims to act on this gland, the pair is where it would show and the pair is what has never been published for it.
- Prediction 2 — Vitamin D (25-Hydroxy). should be in the 30-100 ng/mL sufficient band before PTH is interpreted at all, 8-12 weeks after a dose change. This is the prediction that will save most readers the price of the product. Deficiency is under 20 ng/mL and insufficiency 20-29, both are common, and both raise PTH as a normal physiological response. A high PTH in a vitamin D-deficient person is usually a vitamin D problem wearing a parathyroid mask, and correcting D is the intervention with the evidence.
- Prediction 3 — Magnesium, RBC. worth checking at 4.0-6.4 mg/dL, because PTH secretion depends on magnesium, 12 weeks after starting supplementation. This is the deep cut and it is real, named physiology rather than speculation: severe magnesium depletion impairs PTH secretion and blunts its action, producing a low-calcium state that does not correct until magnesium does. Red-cell magnesium reflects stores far better than a serum value. It is a cheap test almost nobody in this market mentions.
- Prediction 4 — calcitonin. is NOT a parathyroid test; under 8.4 pg/mL is the band and it belongs to the thyroid, only if an endocrinologist asks for it. This entry is here as a boundary rather than a prediction. Calcitonin comes from thyroid C-cells and is a tumor marker for medullary thyroid carcinoma. Confusing the thyroid and parathyroid glands is common enough that a page about one should say plainly which tests belong to which.
Run these before and after, not after alone. A single post-course number tells you what your body is doing, not what Bonothyrk did to it — and that difference is the entire point of testing.
Bonothyrk versus the alternatives
Bonothyrk versus the thing that actually raises PTH in most people. This is the comparison worth the price of the page. A raised PTH with a normal or low calcium is, far more often than not, secondary hyperparathyroidism driven by vitamin D deficiency — deficient is under 20 ng/mL and insufficient 20-29, against a sufficient band of 30-100 — and the treatment is to correct the vitamin D and retest in 8-12 weeks. It is cheap, it has an enormous evidence base, and it makes the PTH come down for a reason anyone can explain. A capsule with 0 published results is not competing with that.
And the second alternative nobody mentions: magnesium. PTH secretion and PTH action are both magnesium-dependent, and severe magnesium depletion produces a functional hypoparathyroid state that does not correct until magnesium does. Red-cell magnesium, banded at 4.0-6.4 mg/dL, reflects stores far better than a serum value. That is real, named physiology with a cheap test attached, and it is a more likely explanation for a strange PTH than anything on this label.
Finally, the one that is not optional. A calcium above 10.2 mg/dL with a PTH that is not suppressed is primary hyperparathyroidism until proven otherwise, and that is a surgical diagnosis with a cure rate most operations would envy. No supplement belongs in that conversation.
What you are actually buying when you buy Bonothyrk
An extract has no identity test. A certificate covers sterility, endotoxin, total protein and named contaminants, and cannot establish contents: a preparation defined by its process has no formula and no mass to confirm, unlike a 3- or 4-residue synthetic where 1 mass-spectrometry run settles it.
The source-tissue question is unusually pointed here. The parathyroids are 4 small glands embedded on the posterior surface of the thyroid, weighing a fraction of a gram apiece, and they are easy to include with thyroid tissue by accident during dissection. A buyer has no way to know which gland a 'parathyroid' preparation actually came from, and no certificate distinguishes them, because neither has a marker a routine certificate reports. That is not an allegation. It is an anatomical fact about what would have to go right in the abattoir for the label to be true, and it is worth knowing before spending anything.
A certificate on this product can establish sterility, endotoxin, total protein and the absence of named contaminants, and cannot establish what it is. The specific point for a parathyroid product is that no certificate tells you the direction of any effect, and direction is the entire clinical question here.

Cell, rodent, human — and where it stops
Nothing has been published on a parathyroid peptide preparation. A PubTator3 search on 6 September 2026 returned no indexed record under this trade name, in any language, and no study of this family with a parathyroid, calcium or bone-density endpoint. Parathyroid tissue is also absent from Ryzhak 2015, the seven-tissue organotypic survey that gives several other products in this catalog their only cell-level result.
What is left is the class argument. Khavinson 2001 and Khavinson 2012 state that an organ's short peptides act preferentially on that organ; Khavinson 2021 is the systematic review of the gene-expression claim; Khavinson 2023 scored 26 ultrashort peptides against LAT and POT carriers by molecular docking and measured no transport in any living cell. None of that is evidence about a parathyroid gland.
The parathyroid is a strange organ to sell an unmeasured product for, because it is defined by a number. The gland exists to hold serum calcium inside a narrow band, and it does so by secreting parathyroid hormone. Both are ordinary, highly standardized blood tests. Unlike a mood, a joint or an energy level, parathyroid function is not a matter of interpretation — it is two values read against a reference range.
Where it stops. Zero published calcium values. Zero PTH values. Zero bone density measurements. For a product named after the gland that governs calcium, that is not a thin evidence base; it is the absence of any attempt.
What nobody has tested yet
Calcium, PTH and 25-hydroxyvitamin D, before and after. Three tests on one panel, drawn together because they are only interpretable together. This is the complete experiment for this product, it costs less than a course, and nobody has published it.
Bone density is the slower, harder endpoint. DEXA at the spine and hip is the standard measure of the outcome a parathyroid product is implicitly sold on, is repeatable at twelve to twenty-four months, and appears nowhere in this class's literature.
Extrapolation, and where it turns into a caution. If a parathyroid preparation raised PTH activity, the predictable consequence is a rise in serum calcium, because that is what the hormone does — and sustained hypercalcemia causes kidney stones, constipation, confusion and bone loss. If instead it suppressed PTH, calcium would fall. Both directions are plausible from the stated mechanism, neither has been measured in any species, and the same single blood test detects both. That is the clearest example in this whole cohort of an unstudied risk that would cost almost nothing to look for.

What the mechanism predicts
Derived from the molecule, not a trial.
- These are short peptide fragments of organ extracts, and the honest starting point is that the mechanism itself is not established in a way that lets anyone predict harm precisely. The proposal is gene-regulatory — short peptides binding DNA and modulating transcription in a tissue-specific way. If that is what they do, the theoretical concern is influencing transcription in tissue you were not aiming at.
- In practice the doses are tiny, the peptides are short, and they are degraded quickly — which is also the argument that they may do very little at all. Those two possibilities are the same uncertainty viewed from opposite ends, and you should hold both.
What has actually been reported
- Decades of Russian clinical use with a strikingly clean tolerability record — injection-site reactions and little else reported.
- That record comes almost entirely from one research school, is largely unreplicated outside it, and safety data from a group with an interest in the outcome is worth less than the same data from a skeptic. This is not an accusation; it is how evidence weighting works.
How to reduce the risk

- Follow the course printed on the label rather than running continuously. These are the one class in the Vault where a duration is STATED rather than inferred, and it is typically 10–20 days repeated a few times a year. Read the box.
- Run one at a time. They are cheap and it is tempting to stack six. If something changes, a stack of six tells you nothing about which one did it — and given the evidence base, attribution is the entire value of your own experiment.
- Decide your endpoint before you start, and make it a marker or a measurable symptom rather than a feeling. With a compound class this under-evidenced, an unfalsifiable endpoint means you will conclude it worked no matter what happened.
- Buy from a source that publishes third-party testing. Where the molecule itself is uncertain, identity and purity are the only variables you can actually control.

- Test the ORGAN, not the peptide. A thymic peptide is judged on immune markers, a pineal one on sleep and IGF-1, a vascular one on lipids and inflammatory markers. There is no assay for the compound itself.
