What actually decides this outcome, in order of size

Six products, one shared obstacle, and the obstacle is about twenty micrometers of dead keratinocytes. Ranked by how much of the outcome each one owns:

1. Whether the peptide crosses the stratum corneum at all. Skin permeability has been called a dismissed necessity for anti-wrinkle peptide performance Mortazavi 2022, which is the most useful single sentence written about this category. The barrier selects strongly against large, charged, hydrophilic molecules, and a hexapeptide is large, charged and hydrophilic. Permeability of collagen pentapeptides and their palmitoylated forms has been measured directly for exactly this reason Choi 2014.
2. How the product is delivered, because delivery is the whole variable. Acetyl hexapeptide-8 has been reviewed specifically for skin permeability and efficacy Zdrada-Nowak 2025, and a cross-linked hyaluronic acid microneedle patch carrying the same peptide has been tested An 2019. A microneedle bypasses the barrier; a cream asks it politely. Palmitoylation exists for the same reason, adding a lipid tail so the peptide behaves like something the barrier will let through Abu Samah 2011.
3. What the honest comparator does, because everybody on this page is comparing themselves to it. Botulinum toxin type A in glabellar frown lines has a dose-response literature Kaufman-Janette 2021. Injections place the molecule at the neuromuscular junction directly. Any topical claiming the same mechanism is claiming to reach the same site through skin, and the size of the gap is the size of the claim.
4. Which line you are treating, because dynamic and static lines are different tissue problems. A dynamic line appears on contraction and is a muscle problem. A static line is visible at rest and is a matrix problem. Only the first is a neuromodulation target, and a peptide aimed at the junction has nothing to offer the second. That distinction decides which half of this page applies.
5. The products, last, and the analytical picture is honest about itself. Argireline has been written up explicitly as an analytical challenge Kluczyk 2021, meaning the compound is hard to detect and quantify where it is supposed to be acting. Instrumented wrinkle measurement of an argireline-containing serum has been reported Henseler 2023, and a serum combining acetyl hexapeptide-8 with a dipeptide and gluconolactone has been evaluated more recently Zhu 2026.

The order to run these in, and what has to be true first

Decide which line you have, treat the matrix that sets its depth, and judge the neuromodulator on a photograph taken under conditions you control. Most of the disappointment on this page is a lighting problem.

1. Separate the dynamic line from the static one, in a mirror, before buying. Frown, then relax. A line that vanishes at rest is the neuromodulation target. A line still visible at rest has a matrix component that Collagen synthesis & dermal matrix is the page for, and no peptide at the junction touches it.
2. Matrixyl belongs first for most readers, because dermal thickness sets how deeply a repeated fold sets. Topically applied KTTKS has been reviewed Abu Samah 2011, the palmitoylated pentapeptide improved photoaged human skin in a controlled study Robinson 2005, and the palmitoyl oligopeptide class has a safety assessment CIR Expert Panel (Cosmetic Ingredient Review) 2012. Its permeation has been measured rather than assumed Choi 2014.
3. Argireline next, with the delivery question asked out loud. Acetyl hexapeptide-8 interferes with SNARE complex assembly, reducing neurotransmitter release at the junction. Its skin permeability and efficacy have been reviewed together Zdrada-Nowak 2025, and it is a recognized analytical challenge to measure Kluczyk 2021. Instrumented wrinkle assessment of a serum containing it exists Henseler 2023.
4. If you are buying this mechanism, buy the delivery. A microneedle patch combining cross-linked hyaluronic acid with acetyl hexapeptide-8 and epidermal growth factor has been tested for anti-wrinkle efficacy An 2019. A formulated serum with a defined combination has been evaluated Zhu 2026. Neither is a generic cream with a peptide on the ingredient list at an undeclared concentration.
5. SNAP-8 and SYN-AKE are the same pathway at two other points, with thinner data. SNAP-8 is an extended analog of the same hexapeptide and inherits the same permeability problem Mortazavi 2022. Syn-Ake acts as a nicotinic acetylcholine receptor antagonist, which is a different point on the same junction and a genuinely interesting mechanism with a thin clinical file. The combination serum evaluated recently included a dipeptide of that class Zhu 2026.
6. Hyaluronic Acid is the fastest visible change on this page and the least structural. Ingested hyaluronan has been shown to moisturize dry skin Kawada 2014 and to relieve wrinkles in a double-blind placebo-controlled study over 12 weeks Oe 2017. Hydration reduces the visible depth of a fine line without changing anything underneath it, which is a real effect and a reversible one.
7. Collagen is the systemic version of the matrix argument and runs on months. Dermal thickness is a slow variable, and the expectation should be set against a 12-week trial rather than a four-week one.
8. Sun exposure outranks everything on this list. It is the dominant modifiable determinant of how a repeated fold becomes a permanent one, it is free, and it belongs at Pigment, tone & photoprotection.

What gets bought for this that cannot move it

The category fails at the stratum corneum, and it fails before pharmacology is relevant. Skin permeability is the necessity this field has dismissed Mortazavi 2022, and the peptides being sold are exactly the molecules the barrier is best at excluding. A peptide that does not reach the neuromuscular junction cannot inhibit SNARE assembly there at any concentration on the label, and the fact that the parent compound is difficult to quantify in the first place Kluczyk 2021 is part of why the question stays open.

SNAP-8 is the option here whose marketing most exceeds its published record. It is sold as a more potent analog at the same target, and the argument for it is structural rather than clinical. Adding residues to a peptide that already struggles to cross the barrier does not obviously improve its odds of getting there.

A topical is not a weaker injection, it is a different route with a different ceiling. The comparator has a dose-response literature at the junction Kaufman-Janette 2021 because it is placed there. Reading a topical result as a fraction of an injectable one assumes both molecules arrive and only the amount differs, and that assumption is the one this page cannot support.

And if the line is not dynamic, this is the wrong page. Lines visible at rest, crepiness and laxity belong at Collagen synthesis & dermal matrix. Uneven tone and sun damage belong at Pigment, tone & photoprotection. Redness, flushing and papules belong at Inflammatory skin — acne, rosacea, eczema, psoriasis. Somebody whose actual question is whether to have an injectable is asking a clinician, not a shelf.

How you would know it was working, on a real read-out and a real timescale

This is a page with no blood test for its own mechanism, so the primary read-out is a photograph and the discipline is in the conditions. The prediction is that a topical neuromodulator should reduce a line depth measured on maximal contraction and should do comparatively little at rest, because that is what the mechanism claims.

• Two photographs at each timepoint, at rest and at maximal contraction, at baseline, 8 weeks and 12 weeks. Both expressions because the dynamic and static components separate only in that comparison. Same camera, same distance, same time of day, same light source and no makeup. Twelve weeks because the instrumented studies in this category report at that order of duration Henseler 2023 Oe 2017.
• HbA1c (Hemoglobin A1c) at each end of the 12 weeks. This is the systemic variable with the most direct route to the tissue: glycation cross-links long-lived dermal collagen, which stiffens the matrix and changes how a repeated fold sets. It is the one number on this page that acts on the static component rather than the dynamic one.
• Vitamin C once. Ascorbate is a required cofactor for the prolyl and lysyl hydroxylases that stabilize the collagen triple helix, so a deficiency puts a ceiling on every matrix intervention on this and the neighboring page. It is genuinely non-optional and genuinely not a treatment.
• TSH (Thyroid-Stimulating Hormone) with Free T4 (Thyroxine) once if skin texture has changed generally. Thyroid status alters dermal glycosaminoglycan content and skin hydration, and it is the systemic imitator most likely to be mistaken for aging in somebody in their forties.
• Vitamin D (25-Hydroxy) read as an exposure marker rather than a target. On this page a rising 25-hydroxyvitamin D in somebody not supplementing is a record of ultraviolet exposure, which is the dominant driver of the static lines the peptides cannot touch. That makes it the one marker here that can be read in the unflattering direction.

What will fool you. A photograph is a lighting condition: side lighting deepens every line and flat frontal lighting removes them, so before-and-after pairs taken in different rooms are worthless. Hydration changes fine-line visibility within hours Kawada 2014, which is why any product containing hyaluronic acid shows an immediate improvement that says nothing about the peptide beside it. Facial swelling on waking flattens lines, so morning and evening photographs are not comparable. And the peptide itself is difficult to measure where it is meant to act Kluczyk 2021 Mortazavi 2022, so there is no confirmation available that the active ingredient ever arrived.
